As for the monofilament thread, I don't think the sheen would ruin it, especially since the turtles are swimming and the whole quilt is really going to be a water scene, right? I am usually matchy-matchy when it comes to thread and appplique, but I truly think you can use the monofilament here. Also, your one block wonders and block party block look fabulous!
